How to examine compatibility and dilution ratio of diluent with Biorelevant Media
Before HPLC analysis of any biorelevant dissolution sample containing drug, it is important to have identified a compatible diluent for the biorelevant medium that will be used. To check the compatibility of diluent with the biorelevant medium, dilute biorelevant medium alone with diluent at various ratios (for example 1:2, 1:5 and 1:10). In most cases, mixtures of methanol and water or buffer are suitable for our biorelevant media. The proportion of methanol to water or buffer will depend upon the individual drug that will be tested in the biorelevant medium. An example of 1:2 dilution ratio is shown below.
