How to prepare the working drug solution containing dosage form (e.g. drug and excipients) with biorelevant medium to check interference
Three steps are required to prepare the working drug solution containing dosage form;
1. Transfer the appropriate amount of the dosage form (prepare same concentration as PSS) into volumetric flask and add organic solvent to dissolve the drug. This will be primary drug solution.
2. Afterwards, prepare the secondary drug solution by diluting the appropriate volume of centrifuged (to remove insoluble excipients) primary drug solution with diluent to prepare a sample with the same drug concentration as SSS.
3. Use biorelevant medium and diluent (optionally) to prepare the working drug solution by diluting the secondary drug solution (which will have the same drug concentration as WSS).
